Description
Follicle-stimulating hormone (FSH) is a glycoprotein gonadotrophins composed of two dissimilar subunits, α and β , joined noncovalently. It is synthesized and secreted by pituitary basophils, with a molecular weight of approximately 33 KD. The α-subunit of FSH is similar to that of the LH and HCG, which composed of 92 amino acids. The β-subunit is unique, composed of 115 amino acids, and distinguishes FSH from the other glycoprotein hormones. FSH receptors (FSH-Rs) are localized to testicular Sertoli cells and ovarian granulosa cells and are coupled to activation of the adenylyl cyclase and other signaling pathways. Activation of FSH-Rs is considered essential for folliculogenesis in the female and spermatogenesis in the male. The peak of FSH is in mid menstruation, but this peak is much lower than that achieved by LH. During menopause, due to ovarian atrophy, estrogen secretion is significantly reduced, thus stimulating the pituitary gland to secrete a large amount of follicular stimulating hormone. Measurement of FSH concentrations can be used to illustrate the dysfunction of the hypothalamic-pituitary-ovarian system.

Follicle Stimulating Hormone Assay Kit is a chemiluminescent microparticle immunoassay (CMIA) for the quantitative determination of Follicle Stimulating Hormone (FSH) in human serum and plasma. The assay kit is intended for in vitro diagnostic use.
Principle
The Follicle Stimulating Hormone assay is a quantitative sandwich immunoassay to determine the presence of FSH in human serum and plasma using CMIA technology with flexible assay protocols.
- Sample, and paramagnetic anti-FSH coated microparticles are mixed, FSH present in the sample binds to anti-FSH coated microparticles, forming an antigen antibody complex.
- After incubation, a conjugate containing acridinium-labeled anti-FSH is added to the reaction mixture and binds to unoccupied binding sites of the anti-FSH coated microparticles.
- After further incubation and washing, Pre-Trigger and Trigger Solutions are added to the reaction mixture.
- The resulting chemiluminescent reaction is measured as relative light units (RLUs). There is a relationship between the amount of FSH in the sample and the RLUs detected by the Optical system. Results are calculated automatically based on the previously established calibration curve.
Storage
The kit should be stored upright, not upside down or horizontally. Store at 2–8°C with a validity period of 12 months.
