Description
Thyroxine (T4) is an iodine-containing hormone secreted by the thyroid gland, which circulates in the blood usually as an equilibrium mixture of free hormone. Free T4 (FT4) is the unbound and biologically active form, which represents only 0.03% of the total T4. This small percentage of the total T4 represents the physiologically available hormone which is biologically active. Once the free T4 is absorbed by the target cells, the equilibrium reestablishes circulating free T4 levels. The remaining T4 is inactive and bound to serum proteins such as thyroxine binding globulin (75%), pre-albumin (15%), and albumin (10%). The determination of free T4 has the advantage of being independent of changes in the concentrations and binding properties of these binding proteins. Therefore, free T4 values may provide the best indication of thyroid dysfunction.

Intended Use
Free Thyroxine Assay Kit is a chemiluminescent microparticle immunoassay (CMIA) for the quantitative determination of Free Thyroxine (FT4) in human serum and plasma. The assay kit is intended for in vitro diagnostic use.
Principle
The Free Thyroxine assay is a quantitative competitive immunoassay to determine the presence of FT4 in human serum and plasma using CMIA technology with flexible assay protocols.
- 1.Sample, and paramagnetic anti-T4 coated microparticles are mixed, T4 present in the sample is displaced from the T4 binding protein and binds to anti-T4 coated microparticles, forming an antigen antibody complex.
- 2.After incubation, a conjugate containing acridinium-labeled T4 is added to the reaction mixture and binds to unoccupied binding sites of the anti-T4 coated microparticles.
- 3.After further incubation and washing, Pre-Trigger and Trigger Solutions are added to the reaction mixture.
- 4.The resulting chemiluminescent reaction is measured as relative light units (RLUs). There is a relationship between the amount of FT4 in the sample and the RLUs detected by the Optical system. Results are calculated automatically based on the previously established calibration curve
Storage
The kit should be stored upright, not upside down or horizontally. Store at 2–8°C with a validity period of 12 months.
