Description
The N-terminal prohormone of brain natriuretic peptide (NT-proBNP) is a prohormone with a 76 amino acid N-terminal inactive protein that is cleaved from the molecule to release brain natriuretic peptide. It is with a relatively long plasma half-life, which would lead to a higher stability of its levels in patients. Measurements of NT-proBNP are used as an independent predictor of congestive heart failure (CHF) and coronary artery disease (CAD) such as acute coronary syndrome (ACS) and stable angina pectoris (SAP). Other potential clinical uses of NT-proBNP are risk stratification of patients with acute coronary syndrome (ACS) and CHF, as well as treatment and monitoring in patients with left ventricular dysfunction (LVD). A significant correlation was observed between BNP and NT-proBNP at all stages of kidney function, but the correlation was significantly affected by kidney function. Observational studies showed a close inverse relationship between plasma NT-proBNP and creatinine clearance. NT-proBNP is also increased in left ventricular hypertrophy and left ventricular systolic and diastolic dysfunction. It is also indicated that an age- and gender difference in the ability of NT-proBNP is considerate to identify myocardial dysfunction in relation to metabolic disturbances.

Intended Use
N-terminal pro B-type natriuretic peptide Assay Kit is a chemiluminescent microparticle immunoassay (CMIA) for the quantitative determination of N-terminal pro B-type natriuretic peptide (NT-proBNP) in human serum and plasma. The assay kit is intended for in vitro diagnostic use.
Principle
The N-terminal pro B-type natriuretic peptide assay is a quantitative sandwich immunoassay to determine the presence of
NT-proBNP in human serum and plasma using CMIA technology with flexible assay protocols.
- Sample, paramagnetic anti-NT-proBNP coated microparticles and a conjugate containing acridinium-labeled anti- NT-proBNP are mixed, NT-proBNP present in the sample binds to anti- NT-proBNP coated microparticles and acridinium-labeled anti- NT-proBNP, forming an antigen antibody complex.
- After further incubation and washing, Pre-Trigger and Trigger Solutions are added to the reaction mixture.
- The resulting chemiluminescent reaction is measured as relative light units (RLUs). There is a relationship between the amount of NT-proBNP in the sample and the RLUs detected by the optical system. Results are calculated automatically based on the previously established calibration curve.
Storage
The kit should be stored upright, not upside down or horizontally. Store at 2–8°C with a validity period of 12 months.
